Materials and Methods: The Ct values for the G and T alleles for two controls (50% and 0.2%) from 20 consecutive clinical runs were used to generate Levey-Jennings charts. We applied the Westgard multi-rules to an in-house developed quantitative JAK2 (c.1849G>T) mutation assay, which uses realtime TaqMan probe technology. These rules consist of a sequence of interrogations and evaluations of output data in accordance with certain decision criteria, usually prompted by control aberrations >2SD from the mean (12S rule). The Westgard multi-rules for QC are currently used in high-throughput clinical laboratories to monitor assay performance over time in order to optimize the detection rate of systematic and random errors. With the emergence of quantitative molecular assays for which the results may be used to guide therapy or where very low mutation levels may be used to make a diagnosis, there is an interest in optimizing QC. Introduction: Commonly used quality control(QC) methods in the molecular laboratory include control charts tracking real-time PCR Ct values within pre-defined, acceptable value ranges, or, for other quantitative assays, monitoring of the standard deviation of sensitivity controls. ![]()
